Self-Reported Ethnicity and Genomic Ancestry in Women of the South of Bahia with Urinary Incontinence: Case - Control Study

Abstract

OBJECTIVE: To estimate genomic ancestry and its association with self-reported ethnicity in samples from women diagnosed with urinary incontinence (UI). METHODOLOGY: 145 women participated, with 52 cases diagnosed by urodynamic evaluation and 93 controls, aged between 45 and 60 years. Six genomic ancestry markers (AIMs) (RB2300, LPL, AT3-I / D, Sb19.3 and APO) were used with frequency differential values δ> 40% between Africans and Europeans or Amerindians. Allele frequencies were analyzed using the GENEPOP program and the proportions of genomic ancestry through the ADMIX 3 program. RESULTS: The prevalence of the self-declared mixed race (69.2%) did not represent a risk factor for UI (p = 0.446) and OR = 0.446 (0.322-1.647). However, the estimate of genomic ancestry assessed by AIMs revealed a greater contribution of European ancestry (66% and 68%), followed by African (24% and 22%) and Amerindian (10%), among the case and control samples. The analysis of self-declared ethnicity compared to genomic ancestry showed disagreement, 20% of women with UI declared themselves white, 80% were brown and black, but the estimate of genomic ancestry showed that the European, African and Amerindian contributions were 66%, 24% and 34%, respectively. CONCLUSION: The difference found between the genomic ancestry obtained using the AIMs and the one verified by the self-declared ethnicity indicate the importance of assessing the degree of ethnic mixture in mixed samples, in order to avoid biases of association with a given disease, as the samples cases and controls may differ significantly in relation to genomic ancestry.
